Heterogeneity and environmental stress resistance of wild yeast isolates
[This dataset is embargoed until August 31, 2020]. This dataset contains analytical results for wild yeast isolates from soil samples at and near a disused metal smelting works in the north-east of the UK. The main contaminant from the smelting works was lead (Pb), but Cd was also elevated. The soil near the smelting works showed above-background levels of Pb. Yeasts of similar colony morphology on laboratory agar were isolated and proved to be mostly isolates of Saitozyma podzolica: 56 independent clonal isolates of this yeast were confirmed by ITS sequencing and RAPD-PCR. All of the S. podzolica isolates were tested for Pb and Cd resistance (IC50) and heteroresistance. Heteroresistance indicated variation in metal resistance between individual cells of clonal populations. Some isolates were also tested with Zn. Soil-core samples were collected during July 2014 near the disused St. Anthonyâs metal smelting works, at either contaminated (16.03 g kg-1 Pb, 10.87 mg Kg-1 Cd) or nearby less-contaminated (4.38 g kg-1 Pb, 1.49 mg Kg-1 Cd) areas. Coordinates and a description of the site are available at http://www.twsitelines.info/SMR/4192. Soil-core samples were diluted in the laboratory and spread to agar media for yeast isolation. Single colonies were subcultured and yeasts identified with ITS sequencing and RAPD PCR. 56 clonal isolates of Saitozyma podzolica were assayed for lead (Pb) and cadmium (Cd) resistance (IC50) and heteroresistance. Some isolates were also tested with zinc (Zn) Genomic DNA preparations submitted to NBAF-Liverpool for whole-genome sequencing. The sequence data were deposited with EBI/NCBI.
