Lineage

A CytoSense fixed beam scanning flow cytometer, fitted with a 488nm blue argon laser was used to measure phytoplankton size and abundance. The flow cytometer was connected to the ferrybox on board the RV Cefas Endeavour with a continuous water intake at approximately 4m depth. Samples were analysed inline for 10 minutes each hour without the addition of a fixative. Data file cleaning (removal of debris) and calibration (using a microsphere of known size, and amicrosphere with strong yellow fluorescence; F8852, Life Technologies, USA) was carried out with CytoClus v3 software (CytoBuoy, the Netherlands).
